My laboratory combined protein purification and phyletic patterns to identify key components of the human tRNA splicing machinery: the RNA-kinase CLP1 within the tRNA splicing endonuclease complex; the tRNA ligase complex with RTCB/HSPC117 as the catalytic subunit; and Archease, which provides multiple turnover to the tRNA ligase. Our dual approach, biochemistry and mouse models, is linking these enzymes to other biological pathways and disease.
